Maturation and Emigration of Single-Positive …

WebbThymic antigen presenting cells provide the appropriate micro-architecture for the maturation of thymocytes, which “sense” the signaling environment via their randomly generated TCRs. Thymic development is characterized by (i) an extremely low success rate, and (ii) the selection of a functional and self-tolerant T cell repertoire. Though the thymus is fully developed before birth, newborns have an essentially empty peripheral immune compartment immediately after birth. Hence, T lymphocytes are not present in the peripheral lymphoid tissues, where naïve, mature lymphocytes are stimulated to respond to pathogens. In order to populate the peripheral system, the thymus increases in size and upregulates its function during the early neonatal period.

Webb1 jan. 2014 · Thymus is now considered the primary immune tissue devoted to the production and the development of T lymphocytes generated from the interactions of haematopoietic stem cells with thymic stromal cells followed by several maturational stages (Table 6.1 ). WebbT cell maturation is under the control of the hormones secreted by the thymic epithelium. These hormones include thymosin α1, thymopoietin, thymopentin, thymosin β4, and thymulin. On hormonal stimulation, most thymocytes express a CD8+ marker but quickly transition into dual positive (CD4+, CD8+) cells.

Webb20 jan. 2024 · During maturation in the thymus, T cells are selected through the binding properties of their antigen-specific T-cell receptor (TCR), through the elimination of both weakly (positive selection) and strongly (negative selection) self-reactive receptors. However, the impact of thymic selection on the TCR repertoire is poorly understood.
